  1. What cell lines can be grown on the Express Bioreactor?
    All commonly used suspension as well as attachment cells have been grown on the Express Bioreactor including hybridomas, CHO cells and HEK 293 cells.
  2. How can the bioreactor provide such high levels of sustained production?
    Two reasons- a) the large surface area sustains a large cell density and b) the design enables each cell which is only a thin layer of media away from air, a high level of efficient gas exchange and therefore there is no oxygen deprivation like in submerged cultures at high density.
  3. Does one run in a batch, fed-batch or perfusion mode?
    The design lends itself to run in any one of the modes described. Media can be slowly re-circulated from a bottle or bag until the death phase and all product collected or by simply exchanging the media bottle/ bag and some desired frequency the culture can be sustained for weeks. Similarly feeding from a feeding container and removing product continuously makes it a perfusion system.
  4. How do you load the cells on the matrix?
    The cell containing media is slowly re-circulated from the bottle on to the matrix and back to the bottle. After a few hours the cells gently get entrapped in the matrix and media clears up.
  5. How is the media consumption when the Express bioreactor is run in a perfusion mode?
    Since the metabolism is more aerobic, less lactic acid is produced and large volumes of media are not needed for pH control like in other cultures. The media consumption has been found to be 4-6X lower than perfusion cultures either in tanks or hollow fiber based bioreactors.
  6. How are the bioreactors sterilized?
    The bioreactor and attached tubing assembly is supplied pre-sterilized and can be set up and run easily in a standard CO2 incubator. It can be disposed after the run is complete.
  7. What is the material of construction of the bioreactor?
    The bioreactor is made out of polycarbonate, the matrix is cellulosic and the tubing is silicone.
  8. Can cells be grown and harvested from the bioreactor?
    Yes, since the cells are not covalently bound they can be removed. The bioreactor chamber is flooded with media and is gently agitated or shaken to dislodge the cells. The media with the cells is then pumped out into a harvest bottle.
  9. How are excess cells or cell debris removed?
    Over time excess cells or cell debris settles to the bottom of the bioreactor as sludge. This can be periodically removed through the sample port via a needleless syringe.
  10. How do you measure pH, cell count and cell viability?
    The sample in the bioreactor enables you to get a sample and measure cell count, cell viability, pH and glucose.
  11. How do you know when to change the media?
    When the glucose level drops below your desired threshold, the media is changed and the product purified.
  12. Can you grow cell lines that have not been suspension adapted?
    Yes, several weeks of suspension or serum free adaptation can be cut off the time line. These cells can be loaded directly on the matrix and the product produced. Serum to serum free transitions can be simply accomplished in a few hours by changing the media bottle and flushing through the bioreactor with serum free media.
  13. Have any other cell lines been grown on the Express bioreactor?
    Fungal, yeast and insect cell lines have been grown successfully.
